The hunk, 46, has been cast in the film which follows a Hollywood fixer Eddie Mannix in the 1950s working to keep a studio's stars in line, reports Deadline.com.
The Coens who will direct and produce under their Mike Zoss Productions banner alongside Eric Fellner and Tim Bevan for Working Title Films, while Universal Pictures has acquired worldwide distribution rights.
Clooney, who last starred in the 'Monuments Men' which he also co-wrote and directed, has previously worked with the Coens on several films including 'O Brother Where Art Thou' and 'Burn After Reading'.
While Brolin, who hit the big screen in 2013's 'Labor Day' which was based on Joyce Maynard's 2009 novel of the same name, last worked with them on 2007's 'No Country for Old Men.'
The American neo-Western thriller saw Brolin play a welder and Vietnam veteran who flees with two million dollars in drug money that he finds in an open field in Texas.
