The actor - who is currently shooting comic book movie 'Green Lantern' - has beaten competition from James McAvoy, Zac Efron, Tom Hardy and Shia LaBeouf for the role in the thriller alongside Denzel Washington, according to The Heat Vision blog.
Daniel Espinosa is directing the project, which tells the tale of a CIA agent who must get a lethal prisoner from one safe house to another without them both getting killed.
Scott Stuber is producing the David Guggenheim script, which was purchased by Universal earlier this year.
Trevor Macy will serve as the executive producer.
Canadian-born Ryan has a number of big projects coming up. As well as 'Green Lantern', where he plays the titular superhero, he has also been announced as the lead character in 'Deadpool' and is due to play a comedy role in David Dobkin's 'The Change-Up'.
